影響MALDI-TOF-MS微生物鑒定的因素有很多,。本研究主要從微生物培養(yǎng)基、培養(yǎng)狀態(tài)、菌體前處理,、上樣量等方面進(jìn)行MALDI-TOF-MS操作技術(shù)和方法的探討。研究結(jié)果顯示:進(jìn)行MALDI-TOF-MS細(xì)菌鑒定,,培養(yǎng)時(shí)間在4~24 h之間為佳,;盡量選擇無(wú)色、不含鹽的液體培養(yǎng)基,;0.1 μL菌液是最低鑒定菌量,;上樣量對(duì)結(jié)果無(wú)影響;對(duì)于菌體前處理,,除按照標(biāo)準(zhǔn)化前處理程序操作外,,也可將菌體加ddH2O制成混懸液進(jìn)行點(diǎn)靶鑒定,這樣更加簡(jiǎn)便,。本研究有助于減少MALDI-TOF-MS操作盲目性,,提高鑒定質(zhì)量,利于操作的規(guī)范化和簡(jiǎn)便化,。
Influence Factors Analysis onMALDI-TOF-MS in Microorganism Identification
Various factors could influence the effectof MALDI-TOF-MS in microorganism identification. In this study,,the approaches and techniques ofMALDI-TOF-MS from aspects of microbiological medium,bacteria incubation state,,bacteria pre-treatment and sample loadingquantity were summarized and discussed. According to the research results,,the period of bacterium incubation of 4 to24 hours was the best;colorless and salt-free medium was preferable,;the lowest ascertainable bacteria solutionwas 0.1 μL,;the sample loading quantity had no effect onidentification result. Besides,for the standard pre-treatment procedures,,a handier way was to add ddH2O into the bacteriato make suspension and conduct targetidentification. This study would contribute to reduce the operational blindnessof MALDI-TOF-MS,,enhance the identification quality andfacilitate the standardization and simplification of operation.
